Polymer-Based Prodrugs for Cancer Chemotherapy

چکیده
Chemotherapy has achieved great success in cancer treatment during recent past decades, but it is still challenged by poor solubility, low tumor selectivity, and associated toxicity of most anticancer drugs. The prodrug strategy is one of the most commonly used chemical/biochemical strategies towards improving the therapeutic index of anticancer drugs. A prodrug is defined as a chemically modified drug derivative that is inactive or less active but metabolized in vivo to release the parent active component in the pharmacological environment to improve the drug’s desirability, including water solubility, patient acceptability (e.g. decreasing pain on injection), and pharmacokinetics (absorption, biodistribution, metabolism, and elimination; ADME). A typical prodrug normally consists of three parts (Figure 11.1): (1) the parent drug exerting therapeutic effects; (2) the chemical linker bridging the parent drug and the modifier; and (3) the modifier endowing the prodrug with

Polymer-Based Prodrugs: Improving Tumor Targeting and the Solubility of Small Molecule Drugs in Cancer Therapy.
The majority of anticancer drugs have poor aqueous solubility, produce adverse effects in healthy tissue, and thus impose major limitations on both clinical efficacy and therapeutic safety of cancer chemotherapy. To help circumvent problems associated with solubility, most cancer drugs are now formulated with co-solubilizers. However, these agents often also introduce severe side effects, there...
متن کاملControlled release of nitric oxide chemotherapy using a nano- sized biodegradable multi-arm polymer
Nitric oxide is a cell signaling molecule that can be a potent inducer of cell death in cancers at elevated concentrations. Nitric oxide molecules are short-lived in vivo; therefore, NO-donating prodrugs have been developed that can deliver NO to tissues at micromolar concentrations. However, NO is also toxic to normal tissues and chronic exposure at low levels can induce tumor growth. We have ...
متن کاملPreparation of acrylic-type derivative of ibuprofen and in vitro evaluation studies of its polymeric prodrugs
Acrylic-type polymeric systems having degradable ester bonds linked to ibuprofen were synthesized and evaluated as materials for drug delivery. Ibuprofen, as a non-steroidal anti-inflammatory drug, was linked to 2-hydroxyethyl methacrylate by activated ester methodology in one-pot procedure. The resulting methacrylic derivative of ibuprofen was copolymerized with 2-hydroxyethyl methacrylate and...
متن کاملAnticancer activity of Doxorubicin conjugated to polymer/carbon based-nanohybrid against MCF-7 breast and HT-29 colon cancer cells
The Cancer is one of the world’s most devastating diseases. Doxorubicin (DOX) is an effective chemotherapeutic drug; however, its toxicity is a significant limitation in therapy. Due to the severe side effects of chemotherapy drugs, scientists have tried to load these drugs in nanocomposites. This paper describes a facile and low cost approach for preparation polymeric biodegradable nanohybrid ...

Prodrugs, the inert derivatives of existing drugs have successfully contributed to the modification of their physicochemical properties. The improved antimicrobial potential due to enhanced lipophilicity of some of the synthesized prodrugs of antibacterial agents by various schemes has already been reported. In the current study, synthesis, characterization, and biological evaluation of some mo...
